Effect of Phytohormone , Temperature , Nitrogen Concentration vs . Potassium and Light Quality on formation of Callus and Organs in Tissue Culture of Chinese Cabbage , Brassica campestris ssp . pekinensis

Abstract
This study was carried out to establish a method of vegetative propagation of Chinese cabbage by in vitro culture of the flower stalk and the leaf tissue. Results obtained were summarized as follows:
1. Callus was well induced from the flower stalk and the leaf tissue, and it well greened and rooted on MS medium containing some levels of NAA and cytokinin. However the shoot was not differentiated in any combination of growth substances in the medium.
2. Temperature treatments to the tissues inoculated on MS and LS media containing NAA 2§·/§¤ and kinetin 10§·/§¤ induced the shoot from the flower stalk segment. One day and 3 days treatments with 35¡É were much effective to inducing the shoot on LS and MS media, respectively.
3. Double concentrations of nitrogen to potassium in the original MS medium also induced the shoot from the flower stalk tissue.
4. Blue and red lights were promotive to inducing the shoot when nitrogen concentration was doubled.
